Description

Description: This assay employs a two-site sandwich ELISA to quantitate CDH3 in samples. An antibody specific for CDH3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CDH3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CDH3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CDH3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Placental cadherin (P-cadherin) is a member of the cadherin gene family. Other members include epithelial cadherin (E-cadherin) and neural cadherin (N-cadherin). A great deal is known about N- and E-cadherin. From the cancer standpoint, loss of E-cadherin in lobular breast cancer and in diffuse gastric cancer is a well-established tumor-initiating or promoting event. N-cadherin expression in mammary cell lines can stabilize fibroblast growth factor, resulting in an invasive phenotype. Until recently, P-cadherin was less studied with respect to cancer. It was originally identified as having an important role in implantation and embryogenesis in mice.
